چکیده

The removal of azo dye Naphtol Blue Black in aqueous media with the catalysis of HRP-dextran conjugate was studied. HRP was purified by affinity chromatography using immobilized Con A. Enzyme-dextran conjugate with 1/10 molar ratio was synthesized using dextran aldehyde derivative (Dextran Mw 75.000 Da) and purified enzyme. The reaction was carried out at pH 3, 4, 5, 6, 7 and 8 in a 25, 30, 35, 40, 45, 50, 60, 70 C temperatured crystal cuvette of UV-Vis spectrophotometer. The bleaching of the dye depending on the time was observed with the absorption spectra recorded regularly for one hour. The best results were obtained at pH 4 and 5 for all temperatures studied.

The Effect of Different Stabilizers on Stability of Horseradish Peroxidase- Bovine Serum Albumin-Aflatoxin B1, a Conjugated Tracer for Detection of Aflatoxin B1 in Immunoassay-Based Methods

Aflatoxins are a group of fungal toxic metabolites, which are contaminated certain food commodities. ELISA is one of the sensitive methods for detection of aflatoxins. Preparation and stabilizing of a proper conjugated tracer for detection of aflatoxins is probably the main step for designing an ELISA method. In current study, different stabilizers were applied to stabilize a newly prepared con...

Visual cortical organization at the single axon level: a beginning.

Single axon analysis of visual cortical connections is an important extension of previous anterograde studies using 3H-amino acids or wheat germ agglutinin-conjugated horseradish peroxidase (WGA-HRP). The higher resolution tracers-Phaseolus vulgaris-leucoagglutinin (PHA-L), biocytin, biotinylated dextran amine (BDA) and dextran-conjugates-have already produced new results, simply by providing i...

Anterograde axonal tracing with the subunit B of cholera toxin: a highly sensitive immunohistochemical protocol for revealing fine axonal morphology in adult and neonatal brains.

We report an improved immunohistochemical protocol for revealing anterograde axonal transport of the subunit B of cholera toxin (CTB) which stains axons and terminals in great detail, so that single axons can be followed over long distances and their arbors reconstructed in their entirety. Our modifications enhance the quality of staining mainly by increasing the penetration of the primary anti...

ELUCIDATION OF pK VALUES FOR ACTIVE SITE OF HORSERADISH PEROXIDASE AND BINDING STUDY OF INTERACTION WITH N-PHENYL BENZHYDROXAMIC ACID USING A SPECIAL DIFFERENCE SPECTROPHOTOMETRIC TECHNIQUE

The binding behavior of a competitive inhibitor, N-phenylbenzhydroxamic acid (BHA) against horseradish peroxidase (HRP) was studied in order to understand and predict the interaction mechanism of hydrogen donors with the enzyme. The dissociation constants of the complexes of HRP-BHA, HRP-donor and HRP-BHA-azide were estimated at specified conditions by difference spectroscopy. The binding s...
